Abstract   We conducted a pase II trial of 5-fluoruracil(S-FU), etoposide(VP-l6) and cisplatin in previously untreated patients with metastatic or recurrent gastric adenocarcinoma. Thirty-six patients entered on study from December 1989 to July 1991. The treatment regi- men consisted of a 12 hr continuous infusion of 5-FU(800 mg/m(2)/day) for 5 consecutive days, a bolus iv of VP-16(70mg/m'/day) and cisplatin(25 mg/(2)m/day) for a consecutive days respectively. Treatment was repeated every 3 weeks. Among 30 evaluable patients, 1 patient(3%) achieved a complete response and 9 patients(30%) partial responses, giving an overall response rate of 33%. The median duration of response was 4 months. The median survival for responders was 11 months. Toxicities were common and included bone marrow suppression, nausea and vomiting, alopecia and peripheral neuropathy. In conclusion, a combination chemotherapy with FEP regimen in patients with advanced gastric cancer seems to be comparable to other chemotherapy regimens in efficacy and toxicity.
